Objective: The aim of this study is to design benzofused nitrogen-containing heterocyclic derivatives of mercaptobenzimidazole using molecular docking, synthesis of active derivative and evaluation as potential antimicrobial agent. Materials and Methods: Using an effective procedure, N-substituted mercaptobenzimidazole derivatives were synthesized based on the literature review. The antimicrobial activity of all synthesized compounds was tested against four different organisms: Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans. Molecular docking of mercaptobenzimidazole derivatives against DNA gyrase subunit B Protein Data Bank (PDB): 5l3j and S. aureus tyrosyl-tRNA synthetase PDB: 1jij was performed using docking protocol.
