Multi-unit compressible coated pellets

As conventional immediate release dosage forms have short duration of action, research on modified systems became extensive. Pellets are formulated to prepare oral controlled release dosage forms with gastro-retentive or sustained-release properties. To attain such properties, pellets must be coated and multi-unit compressible tablets must be prepared. They are advantageous in many aspects like masking of taste, avoidance of dose dumping, bioavailability enhancement along with uniform drug absorption. Several factors like nature and amount of polymer, cushioning excipients, core pellets and equipment used, influence the designing of tablets. The polymers used for coating depend upon the type of drug release. The major problem in the preparation of compressible coated pellets is compression. When pellets are compressed, there is a chance for loss of coating. So, the method employed should also preserve coating even after the compression of pellets into tablets. The drug uniformity in pellets is a primary prerequisite, which avoids dose dumping. Blending process, blenders used and the compaction also affects the drug mixing. The current review discusses the advantages, preparation techniques and various factors affecting the formulation of multi-unit compressible coated tablets
